INTERCELLULAR TRANSFER OF p-GLUCURONIDASE IN CHIMERIC MICE
نویسنده
چکیده
Chimeric mice were produced by aggregating 2 embryos, each of which was homozygous for a different structural allele of the enzyme /?-glucuronidase. The two alleles used were Gus, the 'wild-type' allele, and Gus, an allele whose gene product shows decreased activity in all tissues as well as decreased heat stability. Staining of untreated adult chimeric livers for glucuronidase activity revealed a mosaic of high (Gus) and low (Gus*) activity cells. The boundaries between cells of different activity were sharp and revealed no diffusion of enzyme or reaction product. Treating sections at 73 °C before staining led to a decay of staining activity in normal (non-chimeric) Gus/Gus and Giis/Gus tissue. The rates of activity loss under the conditions used differed by 10-fold. Of the 2 genotypes in the chimera, the dark-staining, Gus, cells decayed in a fashion similar to that of the Gus/Gus control. The light-staining, Gus, cells of the chimera lost their staining in a unique fashion. Within 20 min they quickly lost a majority of their staining activity but that which remained was relatively heat stable. The second GusMike phase of the decay, seen both photographically and photometrically, suggests that Giis* gene product has been transferred to cells of Gus/Giu genotype.

Induction of Specific Humoral Immune Response in Mice against a Pseudomonas aeruginosa Chimeric PilQ/PilA Protein
Background: Pseudomonas aeruginosa, an opportunistic pathogen, is a common cause of healthcare-associated infections in immunocompromised individuals. The rapid emergence of multidrug-resistant strains has made P. aeruginosa infections progressively difficult to treat. In this study we evaluated the effect of a chimeric protein containing a P. aeruginosa PilQ fragment and the PilA disulfide loo...
متن کاملDesigning and Expression of Recombinant Chimeric Protein Containing CtxB and OmpW from Vibrio Cholerae and Evaluation of Its Immunogenicity
Background: Cholera disease caused by Vibrio cholerae remains a major cause of morbidity and mortality throughout the world. Various strategies with different proteins as immunogens have been tried for vaccine development, none of which have been sufficiently effective to preclude cholera. Chimeric proteins, with their ability to present multiple antigens at the same time, can play important ro...
متن کاملExpression, purification, and immunization of a chimeric protein containing immunogenic regions of flagellin and intimin proteins against E. coli O157: H7
Introduction: Enterohemorrhagic Escherichia coli (EHEC) and serotype O157: H7 is one of the most important diseases causing diarrhea. Shiga-like toxin secreted by the bacteria destroys epithelial cells and, in acute cases, causes hemolytic uremic syndrome (HUS). Antibiotic therapy is not effective against this pathogen, because it increases the production of Shiga toxin. Designing chimeric immu...
متن کاملEvaluation of immune response to recombinant Bacillus anthracis LFD1-PA4 chimeric protein
Background: Anthrax is a particularly dangerous infectious disease that affects humans and livestock. Efficacious vaccines that can rapidly induce a long-term immune response are required to prevent anthrax infection in humans. Domains 4 and 1 of the protective antigen (PA) and lethal factor (LF), respectively, have very high antigenic properties. Aims: In this...
متن کاملImmunogenicity of chimeric MUC1-HER2 vaccine against breast cancer in mice
Objective(s): Breast cancer is one of the most common cancers in the world and is on the increase. MUC1 and HER2 as tumor-associated antigens (TAAs) are abnormally expressed to some extent in 75–80% of breast cancers. In our present research, a novel chimeric MUC1-HER2 (HM) protein was designed and used to study whether an immune response can be generated against these TAAs. In vitro analysis ...
